Let EcoArray submit your clones to our sequencing facility. Clones are selected and grown in LB broth with glycerol and ampicillin. RCAs are sequenced in the MegaBace sequencer. Sequences will be quality trimmed (prd files required). Vector sequences will also be identified and removed. Additional sequences (i.e., adapters used for subtractive hybridizations) can also be trimmed as long as the sequence information is provided. Occasionally, it is not possible to provide sequence data for a particular clone due to poor growth, bad phred values, or because the entire clone was vector. A single file of the quality trimmed, vector removed sequences will be sent to customer.
Cost: $650 per plate. Sequences must be requested in batches of 96 (to fill one plate).
SEQUENCE ANALYSIS PACKAGE.This package takes your genes several additional steps and includes the following

Assembly of sequences Description. Multiple reads of the same sequence will be identified and assembled to form a continuous sequence (contig). These contigs, as well as unique singlets (sequences that do not match any other sequence in the sequence pool), will be queried (BLAST-X) to the National Center for Biotechnology Information (NCBI) gene databases for gene identification.
Input of sequences into an interactive website Description.The BLAST results for the singlets and contigs will be uploaded into a secure, password protected interactive website called Blast Quest. This searchable website will contain pertinent information obtained from blast results such as expectation (e) values, bit scores, accession numbers, and links to sequence information at PubMed, NCBI.
Sequence output Description.Excel files listing the top hit, accession number, e-value, and bit score for each gene will be generated and sent to users. This information can be incorporated with microarray data to link gene expression levels with gene ID information. Additional files sent to user will also include excel files of the reads that make up a contig. This information is useful in the design of cDNA arrays.
Cost: $750 for each batch of up to 500 clones. This package can be added to all library construction methods or run separately on sequence data that you supply.
